DC Comics' chief creative officer Geoff Johns recently suggested that an adaptation of the comic, created by writer Brian Azzarello and artist Eduardo Risso, was in the works.
The series, which ran from 1999 to 2009, followed the mysterious Agent Graves as he approaches victims of wrongdoing and offers them the opportunity to covertly murder their enemies.
Goyer has previously worked on comic adaptations including the Blade trilogy, Christopher Nolan's Batman trilogy and the Ghost Rider films.
He also directed 2009 horror film The Unborn and served as executive producer on ABC drama FlashForward.
Warner Bros. Television will produce the 100 Bullets pilot for Showtime.
